These controls were conducted to evaluate the potential effects due to conductivity alone. Protocol Used: EPA/821/R-02/013, 2002 Freshwater Chronic Test Acceptability Criteria: The Lab Control must meet or exceed a mean of 80% survival; 60% of the surviving females must produce 3 or more broods of offspring; and surviving females must produce a mean of 15 or more offspring (only the first 3 broods are used for calculating mean reproduction). There were no adverse effects observed with the survival of the test organisms in any of the four sample sites tested. There was, however, an adverse effect or toxicity observed in all four samples with regards to the reproduction endpoint. The adverse effects to reproduction were expected however, due to the elevated levels of conductivity found in the samples. Through internal studies and experience, we have consistently seen that adverse effects begin to show in the reproduction endpoint when conductivity levels exceed 2,500 pS/cm. Conductivity levels are determined by the varying concentration of salts in a sample. The optimum range for Ceriodaptinia is between approximately 100 and 1,500 pS/cm. The correlation between higher conductivity levels and decreased reproduction in Ceriodaptinia was observed in this study as well. Since all four samples had conductivity levels greater than 2,500 pS/cm (ranging from 3,000 to 4,900 pS/cm), an adverse effect to the reproduction was expected. The Well # 3 sample had the highest conductivity level (4,900 pS/cm), and as expected, showed the greatest decrease in the number of offspring produced (an 84% reduction compared to the control). Along with each sample tested, a Conductivity Control was tested. The Conductivity Control was standard laboratory water with salts added to raise the conductivity level to match that of the 100% sample. Each Conductivity Control, with its corresponding 100% sample, resulted in similar reductions in the reproduction rate. The results of this study indicate that the elevated conductivity levels found in the samples may have contributed to the adverse effects observed in reproduction of the Ceriodaphnia test species.
